We use lasers which employ the gaseous chemicals helium, nitrogen, xenon, and HCl to produce light. This light pumps another laser which employs methanol dissolved dyes to produce different colors of light. This final color is used to probe the molecule acetaldehyde which is flowing through a helium matrix. None of these chemicals are open to the air (all are contained within containers or a sample cell or a vacuum chamber).

We use a 1:1 mixture of 48% hydrogen fluoride:Ethanol to etch crystalline silicon wafers in an electrochemical cell. We then use various solvents (the ones listed above) as washes. The alkynes and such are chemically bonded to the surface using the catalyst Ethylaluminum dichloride.

My only interaction with chemicals is through the labs that I instruct. Although many of the components are common household solutions, such as mouthwash, we do use smaller quantities of lye (conc. NaOH soln.), acids that are very caustic (HCl, nitric and sulfuric) and some dilute solutions of heavy metals.

Use different chemical solutions while working with human red blood cells and performing HPLC analyses. Most of the solutions I use are aqueous and non-hazardous. To quantitate the amount of hemoglobin in my samples use drabkins reagent which contains potassium cyanide and ferricyanide. Other than those reagents the most common chemicals I use are solvents such as methanol and ethanol for HPLC.

I have not worked with chemicals for 10 years, but I supervise roughly 20 people who do. The people in my laboratory work primarily with biochemicals. There work with liquid chromatography causes them to work with liter quantities of methanol and acetonitrile. They also use small quantities of trifluoroacetic acid in this work. I use a 95:5 sol of benzene in methanol to dissolve several types of lipids. I hydrolyze these lipids with the .1M and .001M hydrochloric acid. To stop the hydrolysis of the lipids at different time intervals, I freeze the reactions in liqiud nitrogen. During HPLC runs, I use methanol and water in a ratio from 100:0 to 60:40.

I modify glucose or glucose derivatives to be able to form di-, tetra- or octasaccharides that might have clinical properties in the treatment of cancer or other diseases. My research basically involves the planned protection (most of the time in an orthogonal manner) of all the hydroxyl (or amino) groups of glucopyranose, in order to later form regiospecifical bonds with other units (ie, acceptor-donor reactions), and then the removal of the protective groups once the oligosaccharide is formed.

Work with chemicals is rather sporadic. I may culture cells, freeze them back or thaw new supplies. I may prepare media and/or buffers, make cell preps, stain cell preps. i deal with the lab hazardous chemical wastes, and on occasion may procees a waste for disposal following instructions from REM. I order chemicals/reagents, and handle them upon arrival. I can come into contact with small quantities of a wide variety of reagents, chemicals, and drugs, some of which are highly toxic, carcinogenic, or otherwise dangerous.

Preservation of animal tissues and histological processing of these tissues. Preservatives include buffered solutions of 1- 4% paraformaldehyde. Tissues are stained with natural dyes, histochemical and immunochemical reactions. Biological tissues are embedded in resins to be hard enough to be cut into thin sections and to be viewed with the light microscope and the electron microscope.

Being a cell culture labs we work with cell culture media and the biological additives such a serum, insulin etc. We run electrophoresis with acrylamide and agarose (staining the DNA with EtBr), making the solutions and gels ourselves. We stain cells on cover slips fixing them with a dilute solution of formaldehyde. We use different detergents for lysing cells, washing western blots on cellulose nitrate membranes. Our primary and secondary antibodies used in westerns might contain small amounts (.01%) NaN3. We use Liquid N2 for long term storage of cell cultures in 10%DMSO.

As a cell culture lab we work with culture media and the biological additives such a serum, insulin etc. We run electrophoresis with acrylamide and agarose (staining the DNA with EtBr), making the solutions and gels ourselves. We stain cells on cover slips fixing them with a dilute solution of formaldehyde. We use different detergents for lysing cells, washing western blots on cellulose nitrate membranes. Our primary and secondary antibodies used in westerns might contain small amounts (.01%) NaN3. We use Liquid N2 for long term storage of cell cultures in 10%DMSO.

Primary areas of research in our laboratory that utilize hazardous chemicals are electrophoresis and cell culture. We perform protein gel electorphoresis (SDS-PAGE), which utilizes acrylamide, bis-acrylamide, ammonium persulfate, sodium dodecylsulfate (SDS) and TEMED as components of the gel matrix. We also perform DNA electrophoresis, which utilizes ethidium bromide as a DNA stain. Our cell culture work involves both mammalian cells and bacterial cells (laboratory strains of E. coli). The chemical hazards involved in cell culture primarily involve media components, such as antibiotics or other drug selection agents (ampicillin, kanamycin, geneticin/G418, puromycin, zeocin, penicillin and streptomycin) and growth supplements such as epidermal growth factor, insulin and hydrocortisone. In addition, we periodically use the protein synthesis inhibitor cyclohexamide. Lastly, many commercial and laboratory-generated antibody solutions contain 0.01% thimerasol or 0.02% sodium azide as a preservative

Western blot analysis (for protein identification) requires the use of electrophoresis and SDS-Page gels made from hazardous chemicals when in liquid form (SDS, Temed, acrylamide). Preservatives for WB use sodium azide. Our lab also is involved with tissue culture of immortal cancer cell lines which grow in media which can contain components such as antibiotics, ampicillin, and penicillin.

Chenicals are used to prepare buffers necessary to perform cell fractionation and purify cellular components including DNA, RNA and proteins. They are also used in solutions that enable to treat living cells in culture and alter their behavior (chemicals that induce biochemical alteration: e.g. phosphorylation, acetylation of proteins, methylation of DNA). My involvement with hazardous chemicals is minimal. My main job is performing mammalian surgical procedures, and work in embryology and developmental biology. I do administer pre-prepared drugs which are in vials. Weekly I mix various solutions of polyethylene glycol. To prepare tissue for observation by electron microscopy. The tissue is fixed (gluteraldehyde/paraformaldehyde), rinsed in buffer, post-fixed (osmium tetroxide), washed again in buffer, dehydrated through a graded series of ethanol solutions, then Propylene Oxide, then infiltrated with the resin of choice (Epoxy, Spurrs, or LR White).
Sections are cut using an ultramicrotome, stained with heavy metals (lead, uranyl acetete), and observed with the electron microscope.

Primary areas of research in our laboratory that utilize hazardous chemicals are electrophoresis and cell culture. We perform protein gel electrophoresis (SDS-PAGE), which utilizes acrylamide, bis-acrylamide, ammonium persulfate, sodium dodecylsulfate (SDS), and TEMED as components of the gel matrix. We also perform DNA electrophoresis, which utilizes ethidium bromide as a DNA stain. Our cell culture work involves both mammalian cells and bacterial cells (laboratory strain E. coli). The chemical hazards involved in cell culture primarily involve media components, such as antibiotics or other drug selection agents (ampicillin, kanamycin, geneticin/G418, puromycin, zeocin, penicillin, and streptomycin) and growth supplements such as epidermal growth factor, insulin, and hydrocortisone. In addition, we periodically use protein synthesis inhibitor cycloheximide. Lastly, many commercial and laboratory-generated antibody solutions contain 0.01% thimerasol or 0.02% sodium azide as a preservative.

I need to polish the aluminum alloys surface. Etching is not necessary for my work; therefore, there may be no chemicals used during processing. But sometimes before polishing, I may use the acetone and then methanol to remove any oil, grease, or dirt from the sample surface.
