Nearly all the reactions I do take place in the dry box which is a nitrogen filled box with arm lenght gloves as the only point of entry. This keeps the chemicals free from exposure to air as they are all air and moisture sensitive. All products are kept in solvent sealed flasks and the main work I do out of the bow involves using these solvent sealed flasks and putting them under vacuum filtration.

I am studying the synthesis and catalytic activity of ligands on a solid support matrix. The synthesis involves the coupling of the Quinine and Dihydroquine free bases to the 1,4 Dichlorophtalazine. Chemicals are used to prepare buffers necessary to perform cell fractionation and purify cellular components including DNA, RNA, and proteins. They are also used in solutions that enable to treat living cells in culture and alter their behavior (chemicals that induce biochemical alteration, ie: phosphorylation, acetylation of proteins, methylation of DNA). Since I am working in the immunotherapy project and, so far, all my experiments were "in vivo", I didn't work with hazardous chemicals. I used o-phenylenediamine dihydrochloride (OPD) as peroxidase substrat when I measured the antibody titer using ELISA technique. I wore gloves and lab coat when I worked with OPD and I disposed in a proper hazardous waste container.

EtOH is one of chemicals use to extract plant DNA. Sionce a special machine does the larger part of the extraction, I do not contact with EtOH directly, I just need occasionaly to chahnge the bottle in the mashine.

I use small quantities of ehidium bromide to stain agarose gel for DNA visualization. I wear glowes during any contact with agarose containing ethidium bromide. The procedure takes place in a specially designated room. Used agaroge and glowes are thown into hazardous waste before leaving the room.

I wash dishes with soapy water. I then put them in a dishwasher and add dishwasher soap. After that I rinse with deionized water. Sometimes if the beaker has been written on with a sharpie marker I use a little acetone to get it off. I soak my pipetts in deionized water with trace amounts of sulfuric acid in it.
