I grow bacterial cells and force them to overexpress membrane proteins. These proteins are purified by Ni-NTA chromatography and the purity is checked by SDS-PAGE analysis. These samples are further examined using analytical ultracentrifugation. Soon I will test the binding of C14 Ribose to these proteins.

I make solutions using chemicals that may be flammable (eg. ethanols), hazardous (eg., Xylenes), toxic (eg., paraformaldehyde), usually in relatively small quantities (one liter volumes) and rather dilute solutions (biologically compatible with tissue samples). I use a cryostat and store tissue samples in liquid nitrogen or -80oC freezers. I use the autoclave. I use open flames on the bench for sterilization. I heat solutions, usually no hotter than 65oC on the bench or in waterbaths.

We generally use polyacrylamide gels for the analysis of proteins. Starting with 30% of acrylamide gel stock which contains 30% acrylamide and 1% of bis-acrylamide, we adjust the final concentration of acrylamide and add ammonium persulfate and TEMED for solidifying reagents just before we pour the gel to the gel apparatus.

As a biologist, I work with ethanol to disinfect surfaces and also to precipitate DNA. Phenol, chloroform and an isopropanol solution saturated with cesium chloride are also used in DNA preparation. Several gel types are used in the lab: agarose and acrylamide. Acrylamide is the more dangerous of the two. In powder form we use a mask to prevent inhalation of acrylamide. In the destaining solution to visualize bands we use methanol. However in the other gel type, agarose, we use ethidium bromide to visualize nucleic acid. The lab equipment I use most are centrifuges, gel running apparatuses, cell hoods, and pipettes.

Preparation of concentrated viral particles samples for cryo or negative stain transmission electron microscopy. Negative stain samples require uranyl acetate to be applied to EM grids. Cryo-EM requires plunge-freezing in liquid ethane and storage in liquid nitrogen. My work involves crystal structure determination of maltose transporter using X-rays. For this we need highly pure protein samples. The gene expressing the desired protein is overexpressed in E coli. The bacterial cells are later harvested and lysed using French Pressure cell. The membrane fraction is washed and the protein is solubilized using a detergent. For protein purification affinity tags are used and followed by ion exchange or size exclusion chromatograhy.

Most work with chemicals consists of making solutions for use on a mass spectrometer (typically around 10-100 micromolar peptide/protein in a 50/50/1 water/methanol/acetic acid solution, sometimes with a base of some sort instead of acid), usually making at most 1mL of solution. In the lab I work on and assist in the maintenance of several different mass spectrometers, mainly a modified Hitachi 3DQ ion trap, and two modified Finnigan ITMS instruments, as well as a Sciex QSTAR. This also invovles the use and maintanence of a variety of mechanical rough pumps.
