My goal is to perform and optimize the separation of KCl and charged dyes by large scale solution electrophoresis in the CRCR. This would include: Preparing mixtures (containing mixtures of) of boric acid and tris(hydroxymethyl)aminomethane. Here, I will check the pH and conductivity of the various solutions prepared. "There must not be any trace of HCl in the solution of tris(hydroxymethyl)aminomethane when it is made." To one of the solutions, the electrode potentials will be checked before and after electrolysis for 2 hours at various frequencies and voltages. From these reading an Ohm's law plot will be generated. Next, to a selected solution -with their respective concentrations- of boric acid and tris(hydroxymethyl)aminomethane, .001M KCl will be added and electrolyzed for 24 hrs. Determining the steepest gradient, potassium ion and pH distributions in the column. Also checking the solubility of methyl red and thymol blue dyes in the solution. The solution is re-electrolyzed at a predetermined voltage and frequencies to monitor the migration of the dyes. Finally, to study the dependence of the dye separation on voltage, rotation speed and buffer composition to develop an optimum, procedure for the dye separation.
